mRNA-1521 (pan-RAS mRNA vaccine) is a multiepitope neoantigen messenger RNA-based cancer vaccine being developed by RNAimmune. It is designed to target the most prevalent mutations across the three RAS isoforms—KRAS, NRAS, and HRAS—which are found in approximately 25% of all human cancers. The vaccine specifically targets eight common mutations: G12D, G13D, L19F, A59T, G60D, Q61H, K117N, and A146T. By delivering mRNA encoding these specific neoantigens, mRNA-1521 aims to stimulate both T-cell and B-cell immune responses against tumor cells. Preclinical studies have demonstrated that the vaccine can inhibit tumor growth in colon cancer models, with increased efficacy observed when combined with immune checkpoint inhibitors like anti-PD1 antibodies. It is currently being investigated for the treatment of various RAS-mutated solid tumors, including pancreatic, colorectal, and lung cancers.
