An mRNA-encoded trispecific T-cell engager (TCE) designed to target CD19, BCMA, and CD3. Developed by METiS TechBio, this therapeutic utilizes an optimized mRNA sequence formulated in a novel lipid nanoparticle (LNP) to induce broad depletion of B cells and plasma cells. The construct consists of a CD3-binding scFv fused with VHH domains specific for CD19 and BCMA. It is intended for the treatment of B-cell-driven cancers, such as multiple myeloma, and autoimmune diseases like systemic lupus erythematosus (SLE). Preclinical data suggests potent B-cell killing and a favorable safety profile with reduced cytokine release compared to traditional protein-based engagers, particularly when administered subcutaneously. The LNP delivery system is designed for preferential biodistribution to secondary lymphoid organs, including the spleen, bone marrow, and lymph nodes.
