Overview

MT-3724 is a novel engineered toxin body (ETB) designed for the treatment of B-cell malignancies, particularly relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and non-Hodgkin's lymphoma. It consists of an anti-CD20 single-chain variable fragment genetically fused to the Shiga-like Toxin A subunit. This fusion enables MT-3724 to bind specifically to CD20 on B-cells, force internalization into the cell, and induce potent cytotoxicity by irreversibly inactivating ribosomes—leading to apoptosis through inhibition of protein synthesis. Unlike traditional monoclonal antibodies such as rituximab, its mechanism does not rely on immune-mediated cytotoxicity but instead directly kills targeted cells via ribosomal inactivation[1][3][6]. Developed using Molecular Templates' Engineered Toxin Bodies platform, MT-3724 was evaluated in multiple phase I/II clinical trials for patients with relapsed/refractory DLBCL and non-Hodgkin's lymphoma but development has since been discontinued[2][7].
