Overview

MT-3724 + gemcitabine + oxaliplatin is a **combination therapy** evaluated for treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other B-cell non-Hodgkin lymphomas. MT-3724 is an engineered toxin body (ETB) consisting of a CD20-specific single-chain variable fragment (scFv) genetically fused to a modified Shiga-like toxin 1A subunit, designed to selectively target and kill CD20-expressing B cells via internalization and ribosome inactivation, inhibiting protein synthesis and inducing apoptosis. Gemcitabine is a nucleoside analogue that inhibits DNA synthesis, and oxaliplatin is a platinum-based chemotherapeutic that induces DNA cross-linking and apoptosis. The combination has shown high response rates (ORR 85.7%) in early trials, with limitations due to capillary leak syndrome.
