Overview

The mutant K-ras peptide vaccine is an experimental peptide-based cancer immunotherapy developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of non-small cell lung cancer (NSCLC) harboring K-Ras mutations. The vaccine is composed of synthetic peptides that correspond to specific mutations in the K-Ras protein, most commonly at codon 12. It is designed to stimulate the patient's immune system, particularly cytotoxic T lymphocytes (CTLs), to recognize and eliminate tumor cells expressing these mutant neoantigens. In clinical trial protocols, such as the Phase I pilot study NCT00003411, the vaccine is typically administered in combination with sargramostim (GM-CSF), which serves as an adjuvant to enhance the recruitment and activation of antigen-presenting cells, thereby boosting the overall immunogenicity of the vaccine.
