MV049 is an orally bioavailable, small molecule inhibitor specifically targeting the KRAS G12C mutation, developed by Shanghai Bowei Biopharmaceutical Co., Ltd. KRAS is a GTPase that functions as a molecular switch in cell signaling; the G12C mutation (a glycine-to-cysteine substitution at codon 12) locks the protein in a constitutively active, GTP-bound state, driving uncontrolled cell proliferation in various cancers, particularly non-small cell lung cancer (NSCLC) and colorectal cancer. MV049 acts by covalently and irreversibly binding to the cysteine residue in the Switch II pocket of the inactive GDP-bound form of the KRAS G12C protein. This binding traps the protein in its inactive state, thereby blocking downstream oncogenic signaling pathways such as the MAPK/ERK pathway. MV049 is currently in Phase 1 clinical development for the treatment of advanced solid tumors carrying the KRAS G12C mutation.
