Overview
Napabucasin + dexamethasone is a **combination therapy** composed of napabucasin, a small molecule inhibitor that suppresses cancer stemness primarily via inhibition of the STAT3 pathway, and dexamethasone, a synthetic glucocorticoid corticosteroid with wide-ranging anti-inflammatory and immunosuppressive actions. Napabucasin exhibits anticancer activity by inducing cell cycle arrest, apoptosis, and inhibiting proliferation and migration of cancer cells, notably through STAT3 pathway inhibition and potentially ROS generation[1][7][8]. Dexamethasone is used to reduce inflammation and edema, and as an adjunct in oncology for cerebral edema, antiemetic effect, and general anti-inflammatory support[3][6]. This combination has been investigated preclinically (notably in glioblastoma models); however, there is no structured evidence for its clinical development as a marketed or advanced trial combination[1].
