Overview

NAV-006 is a next-generation monoclonal antibody engineered as a "biobetter" version of rituximab. Developed using Navrogen's Block-Removed Immunoglobulin Technology (BRITE), NAV-006 is specifically designed to overcome immunosuppression mediated by the tumor-associated antigen CA125 (also known as MUC16 or HIO-1). In certain lymphomas, high levels of CA125 can bind and suppress the activity of standard anti-CD20 antibodies like rituximab, reducing their efficacy. By removing the CA125 binding site, NAV-006 maintains and enhances immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC) even in the presence of high CA125. Preclinical studies have shown that NAV-006 demonstrates superior antitumor activity compared to parent rituximab in models with elevated CA125/MUC16, suggesting potential clinical benefit for patients with Non-Hodgkin’s Lymphoma and other B-cell lymphomas characterized by high CA125 expression[2][5][7][9][10].
