NCP-CD47 siRNA is an investigational RNA interference (RNAi)-based therapeutic developed by West China Hospital for the treatment of advanced solid tumors. The drug consists of a small interfering RNA (siRNA) targeting CD47, an immune checkpoint molecule widely overexpressed on tumor cells that provides a 'don't eat me' signal to macrophages. This siRNA is encapsulated within a non-cationic peptide (NCP) nanocarrier designed for intratumoral delivery. By silencing CD47 expression in tumor cells, the drug aims to disrupt the SIRPα signaling pathway, thereby restoring immune recognition and promoting the phagocytosis of cancer cells by macrophages and dendritic cells. It is currently being evaluated in a Phase 1, single-arm, dose-escalation clinical trial to assess safety, tolerability, and preliminary antitumor activity in patients with advanced solid tumors refractory to second-line treatment.
