Overview

Necator americanus larvae are the infective third-stage (L3) larval form of the human hookworm parasite Necator americanus. This nematode is a major cause of human hookworm infection worldwide, particularly in tropical and subtropical regions with poor sanitation. The L3 filariform larvae develop in contaminated soil from eggs passed in human feces and become infective after 5–10 days. Upon skin contact—typically through bare feet—the L3 larvae penetrate the skin, migrate via the bloodstream to the lungs, ascend to the pharynx, are swallowed, and mature into adult worms in the small intestine[5][6]. In experimental medicine and research settings, these live infective-stage larvae have been produced under controlled conditions for use in clinical trials investigating their potential as immunomodulatory agents or for studying host-parasite interactions[3]. Mechanistically, N. americanus modulates host immunity during its migration and establishment by producing proteases that degrade connective tissue components such as collagen and elastin[2], facilitating tissue penetration.
