Overview

NGY-A is a small molecule inhibitor of monocarboxylate transporters (MCTs), specifically targeting MCT1 and MCT4. Developed by Nirogyone Therapeutics, NGY-A is designed to modulate the tumor microenvironment (TME) by inhibiting the export of lactate from glycolytic tumor cells. High levels of lactate in the TME are associated with immunosuppression, angiogenesis, and tumor progression. By reducing lactate accumulation, NGY-A aims to restore anti-tumor immunity, particularly by enhancing the activity of cytotoxic CD8+ T-cells. In preclinical studies using melanoma syngeneic mouse models, NGY-A demonstrated significant tumor growth inhibition and downregulation of immunosuppressive markers such as PD-L1, B7-H3, and Galectin-9.
