Novotarg is a bispecific therapeutic antibody being developed by Baliopharm to treat B‑cell malignancies and B‑cell–mediated autoimmune diseases by simultaneously targeting the B‑cell surface antigen CD20 and the apoptosis‑inducing death receptor CD95 (Fas). By binding CD20 on B cells and engaging CD95 only after CD20 recognition, Novotarg is designed to selectively trigger CD95‑mediated apoptosis in CD20‑positive cells while sparing CD95‑sensitive non‑B cells, addressing the off‑target hepatotoxicity and other systemic toxicities seen with monospecific CD95 agonists.[2][10][11] In preclinical in vitro studies, CD20×CD95 bispecific antibodies based on this concept have selectively killed CD20‑positive lymphoma cells, and in mouse xenograft models they markedly suppressed growth of human SKW lymphoma with minimal toxicity to normal CD95‑sensitive cells; additional non‑human primate studies in marmosets have shown proof‑of‑concept for safety and efficacy in models of multiple sclerosis.[2]
