**NPPDT-56MESS** is a non-classical platinum-based chemotherapeutic agent, designed as a potent DNA-intercalating platinum complex. It exhibits highly selective cytotoxic activity against cancer cells, particularly oral cancer and uveal melanoma, and demonstrates superior efficacy compared to cisplatin in preclinical studies. Its mechanism of action includes induction of G2/M phase cell cycle arrest, binding to DNA G-quadruplex structures, downregulation of c-MYC and k-RAS oncogene expression, and triggering of DNA damage. In addition, NPPDT-56MESS activates the cGAS-STING pathway, thereby representing a dual chemotherapeutic and immunomodulatory approach. It does not induce apoptosis directly but inhibits cancer cell proliferation, and overexpression of FACL4 can rescue its growth inhibition effects.
