NPX461 is an investigational cell-penetrating (CPM) fusion protein being developed by Neurophoenix for the treatment of optic neuropathies, specifically non-arteritic anterior ischemic optic neuropathy (NAION) and glaucoma. The drug utilizes a platform based on rabies virus glycoprotein biomimicry to deliver a therapeutic protein that disrupts the protein-protein interaction between Phosphatase and tensin homolog (PTEN) and the kinases MAST1 and MAST2. By inactivating PTEN, NPX461 aims to unlock the intrinsic regenerative capacity of neurons, providing both neuroprotective and neuroregenerative effects to promote the repair and survival of retinal ganglion cells. It is currently in the preclinical candidate stage.
