Overview

NS-229 is an investigational, selective Janus kinase 1 (JAK1) inhibitor being developed for the treatment of eosinophilic granulomatosis with polyangiitis (EGPA), a rare autoimmune vasculitis that affects small to medium-sized blood vessels and can cause tissue and organ damage in the lungs, sinuses, peripheral nerves, skin, and kidneys. By selectively inhibiting JAK1, NS-229 aims to regulate immune cell function and suppress immune-mediated inflammation that leads to tissue damage in EGPA. The drug is currently being evaluated in a global phase 2 randomized, double-blind clinical trial comparing its efficacy and safety against placebo in patients with EGPA who are receiving background corticosteroid therapy. The U.S. FDA has granted Orphan Drug Designation to NS-229 for this indication[1][3][5][8][9].
