NTX-472 is a preclinical, trispecific, mRNA-based immunotherapy developed for the treatment of B cell lymphoma. It encodes a multispecific antibody that simultaneously targets CD20, CD19, and CD47 antigens on B cells. The therapy is delivered as an mRNA nanoparticle formulation that instructs cells to produce the engineered protein in vivo. By engaging these three targets at once, NTX-472 aims to enhance tumor cell killing and overcome resistance mechanisms seen with monospecific antibodies (such as antigen downregulation). Preclinical studies have demonstrated rapid depletion of B cells without detectable binding to red blood cells, suggesting a favorable safety profile compared to other anti-CD47 therapies[2][3][4][6][7].
