Overview

The NY-ESO-1 circRNA vaccine is an investigational circular RNA (circRNA)-based cancer vaccine currently in preclinical development. Developed using the proprietary 'NeoAna' circRNA synthesis platform, the vaccine consists of circular RNA encoding the NY-ESO-1 (New York esophageal squamous cell carcinoma 1) antigen, a highly immunogenic cancer-testis antigen. The circRNA is encapsulated in lipid nanoparticles (LNPs) for delivery. Unlike linear mRNA, circRNA features a covalently closed structure that provides enhanced stability and a prolonged half-life. Preclinical studies in lung cancer models have demonstrated that the vaccine promotes dendritic cell maturation, T-cell activation, and increases the CD8+/CD4+ T-cell ratio. It has also shown synergistic therapeutic potential when combined with anti-PD-1 immune checkpoint inhibitors.
