Overview

Obinutinib is a glycoengineered, type II, anti-CD20 monoclonal antibody designed to treat B-cell malignancies such as chronic lymphocytic leukemia (CLL) and follicular lymphoma. Developed by Glycart Biotechnology (a Roche subsidiary), it targets the CD20 antigen on B cells to induce cell death through enhanced antibody-dependent cellular cytotoxicity (ADCC) and direct apoptosis induction. Unlike type I anti-CD20 antibodies like rituximab, obinutinib's Fc region is afucosylated to increase its affinity for FcyRIIIa receptors on effector cells, thereby boosting its immune-mediated killing capacity. It is frequently used in combination with other targeted therapies, such as venetoclax, or with chemotherapy to achieve deep molecular remissions in patients with hematologic cancers.
