Obinutuzumab + polatuzumab vedotin + lenalidomide is a combination regimen consisting of three active pharmaceuticals: - **Obinutuzumab**: a monoclonal antibody targeting CD20 on B cells, promoting cell death through antibody-dependent cellular cytotoxicity and direct cytotoxicity. - **Polatuzumab vedotin**: an antibody-drug conjugate comprised of a humanized anti-CD79b monoclonal antibody conjugated via a protease-cleavable linker to monomethyl auristatin E (MMAE), a cytotoxic microtubule inhibitor. This triplet is under investigation for the treatment of **relapsed/refractory follicular lymphoma** (FL), leveraging complementary mechanisms for B-cell lymphoma targeting and immune modulation. It demonstrated high rates of complete and objective responses in a heavily pretreated population, with a safety profile consistent with the individual agents. Development has included phase 1b/2 trials at multiple international centers, with induction and maintenance phases for responding patients[1][3][5][6].
