A combination regimen consisting of obinutuzumab (a glycoengineered type II monoclonal antibody targeting CD20), venetoclax (a small molecule BCL2 inhibitor), and lenalidomide (an immunomodulatory agent). This combination is being investigated as a chemotherapy-free approach in patients with follicular lymphoma and other B-cell lymphomas. Obinutuzumab induces cell death primarily through enhanced antibody-dependent cellular cytotoxicity and phagocytosis. Venetoclax selectively inhibits the anti-apoptotic protein BCL2, promoting apoptosis in cancer cells. Lenalidomide modulates immune responses and also induces apoptosis through multiple mechanisms. The regimen aims to leverage complementary mechanisms of action to maximize anti-tumor efficacy and minimize toxicity compared to conventional chemotherapy[1][2].
