**Obinutuzumab + zanubrutinib + pomalidomide + venetoclax** is an investigational combination therapy primarily evaluated for relapsed/refractory chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphomas. Obinutuzumab is a glycoengineered type II anti-CD20 monoclonal antibody that induces direct B-cell killing, antibody-dependent cellular cytotoxicity (ADCC), and phagocytosis via enhanced FcγRIIIa binding. Zanubrutinib is a next-generation BTK inhibitor that covalently binds Bruton's tyrosine kinase to block B-cell receptor signaling. Pomalidomide, an immunomodulatory agent (IMiD), promotes cereblon-mediated degradation of transcription factors like Ikaros and Aiolos, enhancing immune activation and direct tumor cell death. Venetoclax is a selective BCL-2 inhibitor that restores apoptosis in cancer cells by displacing pro-apoptotic proteins. This quadruple regimen synergistically targets CD20+ B-cell malignancies through complementary immune, kinase inhibition, apoptotic, and immunomodulatory mechanisms, showing promising activity in heavily pretreated patients.
