Ocaratuzumab is a humanized Fc-engineered IgG1 monoclonal antibody targeting the CD20 antigen on B lymphocytes. It is designed for enhanced affinity to CD20 and increased antibody-dependent cell-mediated cytotoxicity (ADCC), particularly in patients with low-affinity FcγRIIIa allotypes. Ocaratuzumab demonstrates greater ADCC potency at lower concentrations compared to rituximab and similar activity to obinutuzumab. Its improved efficacy may allow for subcutaneous administration and lower dosing requirements. Ocaratuzumab has been investigated in phase I/II trials for relapsed/refractory follicular lymphoma and rheumatoid arthritis, with ongoing development in other oncology and rheumatology indications[1][2][3][4][5][7].
