Overview

Ofatumumab is a fully human monoclonal antibody that targets the CD20 antigen on the surface of pre-B and mature B lymphocytes. It is used primarily for the treatment of relapsing forms of multiple sclerosis (MS), including clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease, as well as for chronic lymphocytic leukemia (CLL). Ofatumumab works by binding to CD20 on B cells, leading to their depletion through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), phagocytosis, and apoptosis. This reduction in B-cell numbers helps slow disease progression in MS and control malignant B cells in CLL. The drug was originally developed using recombinant DNA technology and has been marketed under the brand names Kesimpta (for MS) and Arzerra (for CLL; now discontinued)[1][2][3][5][6].
