A combination therapy consisting of ofatumumab, a monoclonal antibody, and bortezomib, a proteasome inhibitor. Bortezomib is a reversible inhibitor of the 26S proteasome that blocks the ubiquitin-proteasome pathway, preventing degradation of key proteins and inducing cell cycle arrest and apoptosis. It primarily inhibits the chymotryptic-like activity of the proteasome and suppresses the NF-κB signaling pathway. Ofatumumab is a monoclonal antibody used in treating chronic lymphocytic leukemia and multiple sclerosis.
