Ofatumumab + fresh frozen plasma is a combination therapy investigated primarily for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL). Ofatumumab is a fully human IgG1κ monoclonal antibody that targets the CD20 antigen on B cells, leading to their destruction through mechanisms such as complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. Fresh frozen plasma (FFP) is used in this combination as a source of complement replacement, aiming to enhance the efficacy of monoclonal antibody therapy by replenishing depleted complement proteins necessary for optimal immune-mediated cell killing. This approach has demonstrated tolerability and notable activity in high-risk CLL patients, with minimal adverse events reported[6]. Ofatumumab alone is also approved for relapsing forms of multiple sclerosis and was previously marketed under brand names Kesimpta (for MS) and Arzerra (for CLL)[2][3].
