Ofatumumab + siponimod is an experimental combination of two disease-modifying therapies for multiple sclerosis. Ofatumumab is an anti-CD20 monoclonal antibody that depletes B lymphocytes to reduce autoimmune activity. Siponimod is a selective sphingosine 1-phosphate (S1P) receptor modulator that sequesters lymphocytes in lymph nodes, thus preventing their entry into the central nervous system and reducing inflammation and neurodegeneration. The combination aims to harness dual mechanisms—B cell depletion and S1P receptor functional antagonism—to comprehensively target MS pathophysiology. The combination is being investigated, particularly for pediatric and relapsing forms of multiple sclerosis[1][3][6][7].
