orelabrutinib + bendamustine + rituximab

Orelabrutinib + bendamustine + rituximab is a combination regimen consisting of three agents used in the treatment of B-cell malignancies, particularly lymphomas. Orelabrutinib is a selective Bruton’s tyrosine kinase (BTK) inhibitor that blocks B-cell receptor signaling, leading to inhibition of malignant B-cell proliferation and survival. Bendamustine is an alkylating agent with both alkylator and purine analog properties, causing DNA crosslinking and cell death. Rituximab is a monoclonal antibody targeting CD20 on B lymphocytes, mediating cell lysis through complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. The combination aims to provide synergistic antitumor effects by targeting different mechanisms involved in lymphoma pathogenesis.
