The 'orelabrutinib + Pola-R-CHP regimen' is an investigational combination therapy being evaluated as a first-line treatment for patients with intermediate- to high-risk diffuse large B-cell lymphoma (DLBCL). This multi-agent regimen combines **orelabrutinib** (brand name: Orelabru), a selective, second-generation, covalent small-molecule inhibitor of Bruton's Tyrosine Kinase (BTK), with the **Pola-R-CHP** backbone. Pola-R-CHP consists of **polatuzumab vedotin** (an antibody-drug conjugate targeting CD79b), **rituximab** (an anti-CD20 monoclonal antibody), and the cytotoxic chemotherapy agents **cyclophosphamide**, **doxorubicin**, and **prednisone**. By integrating BTK inhibition with established targeted biologics and chemotherapy, the regimen aims to simultaneously disrupt multiple B-cell survival pathways, including BCR signaling and surface antigens CD79b and CD20, to improve clinical outcomes in previously untreated DLBCL patients.
