Overview

Pro-miniCHOP is a combination chemoimmunotherapy regimen consisting of orelabrutinib, pomalidomide, rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisolone. It is specifically designed for the treatment of treatment-naive elderly patients with diffuse large B-cell lymphoma (DLBCL). The regimen combines the "Pro" triplet—which includes the BTK inhibitor orelabrutinib, the immunomodulatory drug pomalidomide, and the anti-CD20 antibody rituximab—with a dose-reduced "miniCHOP" backbone. This approach aims to enhance therapeutic efficacy by targeting multiple oncogenic pathways and the tumor microenvironment while maintaining a manageable safety profile in an older patient population.
