Overview

This combination therapy is a treatment regimen being evaluated in a Phase 2 clinical trial at Ruijin Hospital for treatment-naive, non-high-risk mantle cell lymphoma (MCL). The protocol consists of induction therapy with **orelabrutinib** (a potent, irreversible Bruton's tyrosine kinase inhibitor developed by InnoCare Pharma) and **rituximab** (a CD20-directed monoclonal antibody), followed by optional autologous stem cell transplantation (ASCT). Patients undergoing ASCT receive the **BEAM** conditioning regimen, which includes the alkylating agents **carmustine** and **melphalan**, the topoisomerase II inhibitor **etoposide**, and the antimetabolite **cytarabine**. The regimen is designed to maximize complete response rates by combining targeted B-cell signaling inhibition with immune-mediated depletion and high-dose chemotherapy-mediated cytoreduction.
