ORIC-1940

ORIC-1940 is an investigational small molecule Janus kinase 1 (JAK1) inhibitor developed by Chengdu Aorui Pharmaceutical for the treatment of hemophagocytic lymphohistiocytosis (HLH). HLH is a life-threatening syndrome of excessive immune activation characterized by a "cytokine storm," where JAK1 signaling plays a critical role in mediating the effects of pro-inflammatory cytokines such as interferon-gamma (IFN-γ). By selectively inhibiting JAK1, ORIC-1940 aims to reduce systemic inflammation and manage the underlying pathophysiology of HLH. The drug is currently undergoing Phase 1 clinical evaluation in China to assess its safety, tolerability, and preliminary efficacy in patients with previously treated HLH.
