Overview

Oxysophocarpine is a bioactive quinolizidine alkaloid primarily extracted from the traditional Chinese medicinal herb *Sophora flavescens* (Kushen). It exhibits a broad spectrum of pharmacological activities, including neuroprotective, anti-viral, anti-convulsant, and anti-nociceptive effects. In oncology research, oxysophocarpine has demonstrated potential against hepatocellular carcinoma (HCC) by inhibiting cell proliferation, inducing apoptosis, and suppressing cell migration. Its mechanism of action involves the downregulation of the **IL-6-mediated JAK2/STAT3 signaling pathway**, which subsequently reduces the expression of **Fibrinogen-like protein 1 (FGL1)**. Since FGL1 is a major ligand for the immune checkpoint **LAG-3**, oxysophocarpine treatment sensitizes HCC cells to CD8+ T cell-mediated immunotherapy, particularly when combined with LAG-3 blockade.
