**P-CD19CD20-ALLO1** is an investigational, off-the-shelf allogeneic dual-CAR T-cell therapy originally developed by Poseida Therapeutics and now being advanced within Roche and Genentech. It consists primarily of donor-derived T stem cell memory T cells engineered to express two full-length CARs with fully human single-domain VH binders directed against CD19 and CD20. The product is designed to eliminate CD19- and/or CD20-expressing B cells and B-cell malignancies, with dual-antigen targeting intended to reduce antigen-escape-mediated relapse. It is manufactured using a nonviral transposon-based engineering approach and incorporates TRBC1/2 and B2M gene knockouts to support allogeneic use. Clinical Phase 1 development includes relapsed or refractory aggressive B-cell lymphomas and multiple sclerosis; a Phase 1 systemic lupus erythematosus study was withdrawn.
