paluratide

Paluratide (LUNA18) is an orally available cyclic peptide and a first-in-class Ras protein inhibitor developed for the treatment of solid tumors. It selectively binds to and inhibits Ras proteins—including KRAS, NRAS, and HRAS—by disrupting the protein-protein interaction between inactive RAS and guanine nucleotide exchange factors (GEFs). This inhibition blocks Ras-dependent signaling pathways that drive tumor cell proliferation in cancers with overexpressed or mutated RAS genes. Paluratide demonstrates oral bioavailability (21–47%) without special formulation and has shown potent activity in preclinical models. The drug is currently in Phase I clinical trials for patients with locally advanced or metastatic solid tumors[1][3][4][5][6][7].
