PAM-194 is a topical small molecule inhibitor of Janus kinase 1 (JAK1) currently under development by Pamplona Biotech (Shenzhen) Co., Ltd. for the treatment of atopic dermatitis. By selectively inhibiting JAK1, PAM-194 modulates the signaling pathways of several key pro-inflammatory cytokines, including IL-4, IL-13, and IL-31, which are central to the pathogenesis of skin inflammation and pruritus in atopic dermatitis. The drug is formulated as an ointment to provide localized therapeutic effects while minimizing systemic exposure and the potential for side effects typically associated with oral JAK inhibitors. PAM-194 is currently being evaluated in Phase 1 clinical trials in China to assess its safety, tolerability, and pharmacokinetics in healthy volunteers.
