A pan-anti-TGF-β antibody is a therapeutic agent designed to neutralize all three mammalian isoforms of Transforming Growth Factor-beta (TGF-β1, TGF-β2, and TGF-β3). TGF-β is a potent pleiotropic cytokine that plays a dual role in cancer, acting as a tumor suppressor in early stages but promoting tumor progression, epithelial-to-mesenchymal transition (EMT), and immune evasion in advanced stages. By targeting all isoforms, these antibodies aim to overcome the redundant functions of the different TGF-β proteins within the tumor microenvironment. In preclinical and clinical settings, pan-TGF-β antibodies are often investigated in combination with radiation therapy or immune checkpoint inhibitors to enhance anti-tumor immune responses and reduce fibrosis. Examples of specific assets in this category include fresolimumab (GC1008) and NIS793.
