Overview

A pan-KRAS small interfering RNA (siRNA) is a therapeutic oligonucleotide designed to silence the expression of the KRAS gene across its various isoforms and mutations. Unlike mutant-selective inhibitors, a pan-KRAS siRNA targets a conserved sequence within the KRAS mRNA to trigger its degradation via the RNA-induced silencing complex (RISC). This approach aims to inhibit the signaling of the KRAS protein, a critical GTPase involved in cell growth and survival that is frequently mutated in pancreatic, colorectal, and lung cancers. In preclinical research, such as that conducted by EnFuego Therapeutics, pan-KRAS siRNAs serve as potent tools to evaluate the impact of total KRAS knockdown compared to mutant-specific approaches like EFTX-G12V.
