PDCD1-edited tumor-infiltrating lymphocytes (TILs) are an investigational autologous cell therapy being developed for the treatment of solid tumors, most notably metastatic melanoma. The therapy involves isolating TILs from a patient's tumor, expanding them in a laboratory setting, and utilizing gene-editing tools—such as CRISPR-Cas9 or zinc finger nucleases (ZFNs)—to knock out the *PDCD1* gene. This gene encodes the PD-1 (Programmed Cell Death Protein 1) receptor, a key immune checkpoint that tumor cells exploit to suppress T-cell activity. By silencing PD-1 expression, the edited TILs are designed to remain active and resistant to immunosuppression within the tumor microenvironment. The treatment regimen typically includes lymphodepleting chemotherapy followed by the infusion of the edited TILs and high-dose interleukin-2 (IL-2) to promote T-cell survival and expansion.
