peficitinib + disease-modifying antirheumatic drugs

Combination therapy involving **peficitinib**—an orally administered small molecule Janus kinase (JAK) inhibitor—and **disease-modifying antirheumatic drugs (DMARDs)** is used primarily for the treatment of rheumatoid arthritis in patients with an inadequate response to conventional therapies. Peficitinib blocks intracellular signaling in immune cells by inhibiting JAK1, JAK2, and JAK3 kinases, thereby suppressing cytokine-mediated inflammatory pathways and reducing synovial inflammation, monocyte chemotaxis, and proliferation of fibroblast-like synoviocytes. The combination with DMARDs augments efficacy in controlling disease progression and joint damage. The therapy is primarily developed and approved in Japan.
