PEG-bPEI nanogel is a non-viral gene delivery platform composed of a polymeric network of polyethylene glycol (PEG) and branched polyethyleneimine (bPEI). Developed by researchers at University College London and the University of Bath, this nanogel system is designed to encapsulate and protect nucleic acids, including plasmid DNA and messenger RNA (mRNA), from enzymatic degradation. The nanogel structure facilitates cellular uptake and intracellular delivery, showing particular efficacy in transfecting difficult-to-reach cell types such as induced pluripotent stem cells (iPSCs) with minimal cytotoxicity. In preclinical studies, it has demonstrated the ability to maintain stable physicochemical properties while enabling measurable gene expression across various cell lines.
