It is created by covalently attaching a 40 kDa polyethylene glycol (PEG) molecule to rhGH (somatropin), which significantly extends its half-life by reducing renal clearance and protecting the protein from enzymatic degradation. This modification allows for once-weekly subcutaneous administration, providing a more convenient alternative to the daily injections required for conventional rhGH therapy. PEG-rhGH acts as an agonist at the growth hormone receptor, triggering the JAK2/STAT5 signaling pathway and stimulating the synthesis and release of insulin-like growth factor 1 (IGF-1).
