Drug intelligence / Profile preview
Pegylated interferon alfa + elebsiran is an investigational combination therapy being evaluated for the treatment of chronic hepatitis B virus (HBV) infection, with the goal of achieving a functional cure. The combination consists of elebsiran (VIR-2218 / BRII-835), a GalNAc-conjugated small interfering RNA (siRNA) that targets a highly conserved region of the HBV genome to degrade viral transcripts and reduce the production of hepatitis B surface antigen (HBsAg), and pegylated interferon alfa (specifically pegylated interferon alfa-2a), a long-acting recombinant interferon that stimulates the host's innate antiviral immune response. By combining siRNA-mediated viral antigen knockdown with interferon-driven immunomodulation, the therapy aims to overcome HBV-induced immune tolerance and restore effective immune control. The combination is being developed by Vir Biotechnology and Brii Biosciences, and has demonstrated promising rates of HBsAg seroclearance in Phase 2 clinical trials.
