This combination therapy consists of the PD-1 checkpoint inhibitor pembrolizumab, the Bruton's tyrosine kinase (BTK) inhibitor ibrutinib, and the anti-CD20 monoclonal antibody rituximab. It is being investigated in a Phase Ib/II clinical trial sponsored by the Dana-Farber Cancer Institute for the treatment of patients with relapsed or refractory primary central nervous system lymphoma (PCNSL). The regimen aims to leverage the synergistic effects of immune checkpoint blockade, targeted inhibition of B-cell receptor signaling, and antibody-dependent cellular cytotoxicity to overcome the blood-brain barrier and treat aggressive CNS lymphomas.
