Overview

PF-06650833 + tofacitinib is an investigational oral combination therapy for the treatment of moderately to severely active rheumatoid arthritis (RA) in patients with inadequate response to methotrexate. PF-06650833 (zimlovisertib) is a highly potent and selective small-molecule inhibitor of interleukin-1 receptor–associated kinase 4 (IRAK4), a key signaling protein involved in innate immune responses and inflammation. Tofacitinib is an approved small-molecule Janus kinase (JAK) inhibitor that blocks JAK1 and JAK3, thereby modulating cytokine signaling pathways implicated in RA pathogenesis. The combination aims to provide additive or synergistic effects by targeting non-overlapping inflammatory pathways. Clinical studies have shown that the combination may offer greater improvement in RA disease activity compared with either agent alone[1][3][7].
