Placebo + rituximab + cyclophosphamide + doxorubicin + vincristine + prednisone is the control arm regimen used in a Phase III clinical trial (NCT04914117) sponsored by InnoCare Pharma. This combination pairs a placebo tablet with the standard-of-care R-CHOP chemoimmunotherapy for the treatment of treatment-naïve patients with the MCD (MYD88/CD79B) genetic subtype of diffuse large B-cell lymphoma (DLBCL). R-CHOP consists of rituximab, a CD20-directed monoclonal antibody; cyclophosphamide, an alkylating agent; doxorubicin, an anthracycline topoisomerase II inhibitor; vincristine, a microtubule inhibitor; and prednisone, a corticosteroid. The study evaluates whether the addition of the BTK inhibitor orelabrutinib to R-CHOP provides superior efficacy over this placebo-controlled standard regimen.
