Overview
Polatuzumab vedotin + rituximab + cytarabine + etoposide + methotrexate is an investigational multi-agent combination chemotherapy and immunotherapy regimen. It is being studied primarily in pediatric, adolescent, and young adult patients with intermediate- or high-risk mature B-cell non-Hodgkin lymphoma (MB-NHL), including Burkitt lymphoma, diffuse large B-cell lymphoma (DLBCL), and high-grade B-cell lymphomas. The regimen combines: - **Polatuzumab vedotin** (an antibody-drug conjugate targeting CD79b on B cells) - **Rituximab** (a monoclonal antibody targeting CD20 on B cells) - **Cytarabine** (a nucleoside analog antimetabolite chemotherapeutic) - **Etoposide** (a topoisomerase II inhibitor chemotherapeutic) - **Methotrexate** (an antimetabolite and antifolate agent) The combination aims to enhance anti-tumor efficacy by leveraging both targeted immunotherapy against malignant B cells and multi-agent cytotoxic chemotherapy. This approach may allow for reduced anthracycline exposure while maintaining disease control[4].
