POLA-Hi-CHP + orelabrutinib is an investigational chemoimmunotherapy combination regimen being evaluated for the treatment of the MCD subtype of diffuse large B-cell lymphoma (DLBCL). This combination targets multiple pathways in B-cell malignancies, including B-cell receptor signaling and direct cytotoxic effects. It is specifically studied in the MCD subtype (defined by MYD88 and CD79B mutations), which often shows sensitivity to BTK inhibition. The regimen is typically administered as induction therapy followed by orelabrutinib monotherapy maintenance.
