Overview
poseltinib + golcadomide + rituximab is an investigational combination regimen being developed for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL). The combination consists of poseltinib, a selective, covalent Bruton's tyrosine kinase (BTK) inhibitor; golcadomide (CC-99282), an oral cereblon E3 ligase modulator (CELMoD) that induces the degradation of transcription factors Ikaros (IKZF1) and Aiolos (IKZF3); and rituximab, a chimeric anti-CD20 monoclonal antibody. This combination leverages the synergistic effects of BTK inhibition, CELMoD-mediated immunomodulation and direct cell killing, and antibody-dependent cellular cytotoxicity (ADCC) to target malignant B cells. It is being evaluated in clinical trials, including the investigator-initiated Phase I/II Go-Pro-DLBCL study.
