The combination of poseltinib, lenalidomide, and rituximab (also known as the R2P regimen) is a chemotherapy-free salvage therapy being developed by NOBO Medicine. This combination is designed to treat B-cell malignancies, specifically relapsed or refractory primary central nervous system lymphoma (R/R PCNSL) and secondary central nervous system lymphoma (R/R SCNSL). Poseltinib is a highly brain-penetrant, selective small molecule inhibitor of Bruton's tyrosine kinase (BTK) and TEC kinase. Lenalidomide is an immunomodulatory drug (IMiD) that targets cereblon (CRBN) to induce the degradation of specific transcription factors. Rituximab is a chimeric monoclonal antibody targeting CD20 on B-cells, promoting cell clearance through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC).
