Overview

PP-P8 is a first-in-class recombinant probiotic-based protein therapeutic developed by Cell Biotech for the treatment of metastatic colorectal cancer. The drug delivery system (DDS) utilizes a genetically engineered strain of *Pediococcus pentosaceus* (CBT-SL4) as a vehicle to deliver and secrete P8, an 8 kDa anticancer protein originally derived from *Lactobacillus rhamnosus* CBT-LR5. Upon oral administration, the probiotic vehicle reaches the intestine and releases the P8 protein, which targets Glycogen Synthase Kinase 3 Beta (GSK3β). P8 exerts its therapeutic effect through a dual mechanism: it binds to GSK3β introns to disrupt transcription and interacts with the GSK3β protein to prevent its phosphorylation-mediated inactivation. This stabilization of active GSK3β promotes the degradation of β-catenin, leading to cell cycle arrest and inhibition of tumor growth. PP-P8 is currently in Phase 1 clinical development for metastatic colorectal cancer, with planned expansions into gastric cancer and diabetes.
